Sudan IV Staining Solution
Harris' Haematoxylin
Acetic Acid 1% Aqueous
Fixation:
Formalin
Sections:
Frozen
Staining Procedures:
- Immerse in 70% Alcohol for an instant.
- Stain for 2 – 5 minutes in Sudan IV Staining Solution.
- Wash quickly in 70% Alcohol. Rinse in distilled water.
- Counterstain in Harris Haematoxylin. Follow with 2 – 5 minutes in Acetic Acid, 1% for clearer blue staining of nuclei.
- Wash thoroughly in tap water.
- Mount (Glycerin or Glycerin Jelly.
Stain Results:
Nuclei | Blue |
Fat | Orange-Red |
Cholesterol | Red |
