Nile Blue A Staining Solution

Sulphuric Acid 1% Aqueous

Fixation

Formalin fixatives

Application

Show differentiation of melanins and lipofuscins

Staining

Deparaffiniz=se and hydrate to distilled water.
Stain 20 minutes in Nile Blue A Staining solution.
Rinse quickly with sulphuric acid, 1%.
Dehydrate immediately in four changes of acetone, 15 seconds each. Rinse in Xylene-acetone. Clear in two changes of Xylene.
Mount.

Stain Results

Lipofuscins	Unstained - natural yellow
Melanins	Green
